When are variable and settings changes applied?

If changes are made in Advanced>DefaultSettings, or Advanced>Variables then does any action need to be taken for FusionPBX to begin using the new information? Thank you.

You may often have to flush memcache too, restarting freeswitch DOES NOT flush memcache.
